P-206: Gestational Diabetes Increased the Astrocytes Density in Cerebellar Cortex of Rat Offspring
Authors
Abstract:
Background: Gestational diabetes mellitus affects 3-5% of all human pregnancy.Studies reported the neurotoxic effect of gestational diabetes on cerebellarand spinal cord in rat offspring. This study was conducted to determine the effect of gestational diabetes on astrocyte density in the cerebellum of rat offspring. Materials and Methods: In this experimental study, 30 Wistar rats dams were randomly allocated in control and diabetic groups. Dams in diabetic group received 40 mg/kg/BW of streptozoto cin, intraperitoneallyat the first day of gestation (GD) and control group received an equivalent volume normal saline. Six offspring of gestational diabetic (GDM) and control dams, at the 21, 28 postnatal days were randomly selected and were sacrificed quickly with anesthesia. The coronal sections of cerebellum (6 micrometer) serially collected. The astrocytes were stained with PTAH. The density of astrocytes was evaluated in 20000 μm2 by OLYSIA Autobioreport software. Results: The astrocyte density in apex anddepth ofcerebellum in 21 postnatal days, in the experimental group significantly increased (38.15 and 25%, respectively) in comparison with controls group (apex:10.5 ± 1.89 vs. 7.6 ± 1.28, depth: 12.25 ± 1.54 vs. 9.8 ± 1.2, p<0.05). The astrocyte density in apex anddepth of cerebellum in 28 postnatal days, in the experimental group significantlyincreased (25% and 13.5%, respectively) in comparison with controls group (apex: 11.25 ± 1.37 vs. 9.0 ± 1.95, depth: 12.25 ± 0.62 vs. 9.75 ± 1.0, p<0.05). Conclusion: This study revealed that gestational diabetes increased theastrocytes density in cerebellar cortex of rat offspring.

Gestational diabetes influences retinal Muller cells in rat's offspring
Objective(s): The Muller cell is the principal glial cell of the vertebrate retina. The expression of Glial fibrillary acidic protein (GFAP) in the Muller cells was used as a cellular marker for retinal damage. This study was done to evaluate the effect of gestational diabetes on retinal Muller cells in rat's offspring. Materials and Methods: In this experimental study, 12 Wistar rat dams were ...
full textThe Effect of Gestational Exposure of Sodium Cromoglycate on Epileptiform Activities in the Rat Offspring
Background: Epilepsy is one of the most common neurological diseases globally. Mast cells are known to be the main source of brain histamine, which is identified as being involved in seizure modulation. Sodium cromoglycate is a popular mast cell stabilizer and is proposed as the first line therapy in chronic asthma and systemic mastocytosis. Considering the importance of gestational period in ...
full textGestational diabetes influences the expression of hypertrophic genes in left ventricle of rat’s offspring
Objective(s): Gestational diabetes increases the risk of congenital heart disease in the offspring, but the molecular mechanism underlying this process remains unclear. Therefore, the current study was conducted to assess the effects of induced gestational diabetes on expression of some involved genes in cardiac hypertrophy in the offspring of diabetic rats.Materials and Methods: Diabetes was i...
full textGestational diabetes leads to down-regulation of CDK4-pRB-E2F1 pathway genes in pancreatic islets of rat offspring
Objective(s): The link between a hyperglycemic intrauterine environment and the development of diabetes later in life has been observed in offspring exposed to gestational diabetes mellitus (GDM), but the underlying mechanisms for this phenomenon are still not clear. Reduced β-cells mass is a determinant in the development of diabetes (type 1 and type 2 diabetes). Some recent studies have provi...
full textGestational diabetes hinders language development in offspring.
BACKGROUND Previous studies have suggested that language is affected in infants of diabetic mothers, yet there have been no systematic investigations to address this question. OBJECTIVE Our goal was to compare infants of diabetic mothers and controls on language outcomes from ages 18 months to 7 years. METHODS This was a case-control longitudinal design with 2 birth cohorts: 1835 singletons...
full textIncreased Cord Blood Betatrophin Levels in the Offspring of Mothers with Gestational Diabetes
AIM Exposing a fetus to hyperglycemia can increase the risk for later-life metabolic disorders. Betatrophin has been proposed as a key regulator of pancreatic beta cell proliferation and lipid regulation. Highly responsive to nutritional signals, serum betatrophin concentrations have been found to be altered by various physiological and pathological conditions. We hypothesized that betatrophin ...
